Tenascin-C is a modular and multifunctional extracellular matrix (ECM) glycoprotein that is exquisitely regulated during embryonic development and in ***** tissue remodeling. In the ***** organism, reexpression of tenascin-C occurs in tumors and other pathologic conditions. The precise function of the protein remains still unknown. Tenascin-C is a hexameric disulphide-linked protein with subunit molecular weights in the range of 210-400 kD. It is primarily made up of 14.5 epidermal growth-factorlike repeat, 15 units similar to fibronectin type III-homology repeat and, at C-terminus, has a sequence with homology to the globular domain of the b and g chain of fibrinogen. In addition, the amino-terminal -150 amino acids form an oligomerization domain allowing interaction of six subunits. Different subunits are generated by alternative splicing of one common primary transcript. Recent reports have suggested that tenascin-C is upregulated during oncogenesis, arthritis, liver, and lung fibrosis. Increased serum levels have been reported in patients with cancer and liver diseases.
