The discovery by Rubanyi’s group in 1985 of a peptide termed endothelium-derived contracting factor (EDCF) and the isolation, sequencing, cloning and naming endothelin by Yanagisawa of the most potent vasoconstrictor has led to more understanding of many physiological effects. ET-1 is known to be one of a family of three mammalian vasoactive peptides including endothelin-2 (ET-2) and endothelin-3 (ET-3). These related peptides differ from ET-1 at the two and six amino acid residue positions, respectively. Those genes make three pre-propeptides that are cleaved into three big endothelin molecules, each 39 amino acid long. A number of reviews of function, molecular biology of the endothelin family of peptides and propeptides are available. ET-1 has been shown to have potent effects on smooth muscle cells, fibroblasts and to be involved in many disease processes, particularly cardiovascular diseases.
