Autoantibodies to pancreatic beta cell antigens are important serological markers of type 1 diabetes mellitus. The antigens recognised by these antibodies include insulin, glutamic acid decarboxylase (GAD65 kDa isoform) and the islet cell antigen IA2 or ICA512. ASSAY PRINCIPLE In DEMEDITEC’s GADAb radioimmunoassay (RIA), GAD antibodies in patient’s sera, calibrators and controls are allowed to interact with 125I labelled GAD65. After a 2 hour incubation, solid phase protein A is added to precipitate labelled GAD-GAD autoantibody complexes. After a further 1 hour incubation period, assay buffer is added to each tube and each tube then centrifuged. Any unbound 125I is removed from the tubes by aspiration of the supernatants. The pellets are then counted on a gamma counter and the % binding determined. The level of radioactivity in the sample is proportional to the amount of GAD antibody in the sample.
