Better hygienic conditions and vaccination caused a decrease of the disease worldwide. Nevertheless every year 400,000 - 800,000 persons die due to this infection. The majority of these persons live in under-developed countries. Protection by vaccination is rare in older persons since Tetanus antitoxin levels decrease with increasing age. Immunity to Tetanus is achieved by vaccination followed by booster injections. Protection is given at a level of 0.1 IU/ml of anti-Tetanus Toxoid. It is advisable to determine the protection status using a qualified test before boostering. Doing this it is possible to minimize effects like local swelling, pain and fever. 3.TEST PRINCIPLE Solid phase enzyme-linked immunosorbent assay (ELISA) based on the sandwich principle. The wells are coated with antigen. Specific antibodies of the sample binding to the antigen coated wells are detected by a secondary enzyme conjugated antibody (E-Ab) specific for human IgG. After the substrate reaction the intensity of the colour developed is proportional to the amount of IgG-specific antibodies detected.
