INTENDED USE
The DEMEDITEC Corticosterone ELISA is an enzyme immunoassay for the quantitative in vitro diagnostic measurement of Corticosterone in serum and plasma.
SUMMARY AND EXPLANATION
Corticosterone is a glucocorticoid secreted by the cortex of the adrenal gland. Corticosterone is produced in response to the stimulation of the adrenal cortex by adrenocorticotropic hormone (ACTH) and is the precursor of aldosterone. Corticosterone is a major indicator of stress since stress increases the production of corticosteroids.
Studies involving corticosterone and levels of stress include impairment of long term memory retrieval, chronic corticosterone elevation due to dietary restrictions and in response to burn injuries. In addition to stress levels, corticosterone is believed to play a decisive role in sleep-wake patterns.
PRINCIPLE OF THE TEST
The DEMEDITEC Corticosterone ELISA Kit is a solid phase enzyme-linked immunosorbent assay (ELISA), based on the principle of competitive binding.
The microtiter wells are coated with a polyclonal antibody directed towards an antigenic site on the Corticosterone molecule.  Endogenous Corticosterone of a patient sample competes with a Corticosterone-horseradish peroxidase conjugate for binding to the coated antibody. After incubation the unbound conjugate is washed off.
The amount of bound peroxidase conjugate is reverse proportional to the concentration of Corticosterone in the sample. After addition of the substrate solution, the intensity of colour developed is reverse proportional to the concentration of Corticosterone in the patient sample.
The polyclonal antiserum of this testkit has been raised in a rabbit. This kit also can be used for any animal sample. In this case we are recommending an extraction procedure as described at the bottom end of this page. As the concentration of corticosterone in rat and mice is about 20 times higher compared to humans this factor has to be taken into concentration when using this testkit for these animals.
Corticosterone is a 21 carbon steroid hormone of the corticosteroid type produced in the cortex of the adrenal glands. In many species, including rats and mice, corticosterone is the principal glucocorticoid, involved in regulation of fuel metabolism, immune reactions, and stress responses. However, in humans, corticosterone is produced primarily in the zona glomerulosa of the adrenal cortex. It has only weak glucocorticoid and mineralocorticoid potencies in humans and is important mainly as an intermediate in the steroidogenic pathway from pregnenolone to aldosterone.

Extraction procedure for steroids
We are recommending an extraction procedure as follows.
Pipette 1 ml of Ethyl Acetate in a 2.5 ml PP microtube (e.g. from Sarstedt).
Add between 5 and 200 µl of your sample.
Mix for 2-3 minutes on a vortexer.
Centrifuge the microtube briefly in order to achieve a clear separation     of the 2 phases.
Deep freeze the microtube at -20°C or lower for several hours (or overnight).
Decant the organic liquid phase into a new tube which can be used for an evaporation process (the specification of the new tube depends of the specifications of your evaporating device). Be sure that no frozen material will be decanted.
Evaporate the organic solvent in an appropriate device at elevated temperature. The boiling point of Ethyl Acetate is 77°C. Please stay clearly below this temperature while evaporating.
After evaporating the organic solvent until complete dryness re-dissolve your sample with a diluent. In most cases this will be the zero standard of the ELISA (or RIA) to be used for the quantification of the analyte.
The volume of the diluent to be used for the reconstitution depends of the sensitivity of your assay system in relation to the analyte concentration in the primary sample.
The dilution (or concentration) factor depends of the volumes you have used in #1 and #7.
