Acid phosphatase (EC 3.1 3.2) (AcP) are a group of enzymes which optimally hydrolyze esters of orthophosphoric acid in the pH range of 4.8 to 6.0. The enzyme has been found throughout tissues and biological fluids. The highest amounts of activity, however, are found in the epithelial cells and secretions of the prostate.
Prostatic acid phosphatase (PAP) enzyme activity was first measured in the urine of men and was found to be localized in organs of the male genital tract. Gutman and co-workers suggested that PAP may be a significant tumor marker in those patients with prostate cancer, because serum PAP concentrations were found to be elevated in many men with primary prostatic carcinoma and metastatic lesions of prostate.
The total serum acid phosphate activity is a representation of pooled enzymes contributed by various tissue cells. Moderate elevation of serum acid phosphatase levels is also observed to accompany many nonprostatic diseases. Therefore, attempts have been made to devise a specific assay for determination of acid phosphatase of postatic origin in the hope of correlating the enzyme level with the progression of prostatic carcinoma. The selection of substrate and inhibitor led to the recommendation of using the tartrate sensitive fraction of serum acid phosphatase detected by 1-naphtyl phosphate to eliminate false positive results derived from erythrocytic and osteoclastic acid phophatase.
However, the colorimetric method cannot distinguish the AcP-2 from the lysosomal acid phosphatase isoenzyme3 (AcP-3) of non-prostatic tissues. This difficulty was overcome by immunochemical methods. The limitation of the polyclonal antibody prepared for AcP-2 is that it cross-reacts with AcP-4 which is present in all tissues. Even some monoclonal antibodies to AcP-2 cannot distinguish these isoenzymes. Therefore, the normal serum values of prostatic Acp-2 includes AcP-4. The next major improvement in the analytical method is to devise a procedure to distinguish AcP-2 from AcP-4.
The DEMEDITEC PAP ELISA provides an enzyme immunoassay for specific recognition of AcP-2. It is a solid phase enzyme linked immunosorbent assay (ELISA). The wells are coated with mouse anti-PAP antibodies. The samples, standards and controls are incubated in the wells together with enzyme conjugate which is a mixture of mouse anti-PAP antibodies with different affinity toward epitopes of PAP molecule and chemically conjugated with horseradish peroxidase. Unbound enzyme conjugate is washed off and the amount of bound peroxidase is proportional to the concentration of the PAP present in the samples, standards and controls.
