性狀：一種水解核糖核酸的酶。商品為白色結晶或凍干粉末,也有50%甘油的稠狀溶液。對濕敏感。極易溶于水。酶反應：專一地催化核糖核酸的核糖部分3′和5′磷酸二酯鍵的裂開,形成具有2′,3′-環(huán)一磷酸衍生物寡聚核苷酸。水溶液在25℃以下相當穩(wěn)定,*大穩(wěn)定范圍pH 2～4.5。*適pH 7.7。*適溫度65℃。等電點(pI)7.8。比旋光度 [α]25D-0.47°(以每mG氮計,c=5,水中)。抑制劑有0.0005mol/L鎂(MG2+)、脫氧核糖核酸(競爭性抑制劑)、變性脫氧核糖核酸(比天然的更有效)。
用途：生化研究,測定核酸的結構,脫氧核糖核酸和核糖核酸的降解。核糖核酸酶 A （Ribonuclease A）來源于牛胰，為高度專一性核酸內切酶，可特異攻擊 RNA 上嘧啶核苷酸的C′3上的磷酸根和相鄰核苷酸的C′5之間的鍵，產(chǎn)物為3′嘧啶單核苷酸或以3′嘧啶核苷酸結尾的低聚核苷酸。可除去 DNA:RNA或RNA:RNA雜合體中未雜交的 RNA 區(qū);可用來確定 DNA 或 RNA 中單堿基突變的位置, 用標記了的RNA探針與待檢含單堿基置換的DNA或RNA退火,RNA：RNA或RNA：DNA中的錯配堿基可被RNaseA切割。廣泛用來去除 DNA 樣品中的 RNA 。
溶液配制：將RNase A溶解在0.001mol/l醋酸鈉(ph5.2)中,使終濃度為10mg/ml,加熱到100°C,15min.室溫下緩慢冷卻,用0.1體積的1M的Tris-Cl調整pH至7.4后,小管分裝保存在-20°C.

Pancreatic RNase A specifically cleaves at the 3′-side of pyrimidine (uracil or cytosine) phosphate bonds. Ribonucleases do not hydrolyze DNA, because the DNA lacks 2′-OH groups essential for the formation of cyclic intermediates. RNase can also hydrolyze RNA from protein samples.
